I would put your servers first, then look at the clients, the DCs will be much quicker than the clients to deal with and you don't need to upgrade the domain or forest function level, unless it's at Rod-IT I had another question we are actually in the middle of setting up a new site and putting a secondary domain controller there and it will be a server R2 server as well.
Do you think I could download the ADMX files from the update and install them on the server R2 and that work with the current primary server and still be able to control our windows 10 computers. I don't see any point in trying, if you are planning on upgrading the domain machines to a newer OS, do that first then it doesn't matter. You cant expect the latest client OS to be supported by a server OS almost 10 years old, the fact Microsoft supported it up to was good of them, and the fact they still support R2 is a blessing for many.
FWIW there is no 'secondary' domain controller, all DCs are equal, and I don't get the logic behind using R2 here if it's a new install you want or at least R2. But then I would have to control the Windows 10 computers from the R2 server though right?

The policy files will synchronize onto all your Domain Controllers and can be centrally managed. GPMC will use the policy files in this location if it exists instead of the local machine.
